dc.description.abstract | Specific Aims: 1) The purpose of this study is to measure and confirm in silico binding affinity of virtual molecular hits to the FAK-FERM domain in vitro.To examine the binding of virtual hits to FAK-FERM for real-time affinity via SPR. 2) To examine the degree of competitive inhibition of FAK-FERM protein-protein interaction with the FAK-FAT domain via virtual hits. Methods: A novel avi-tagged FERM construct was designed, inserted in a plasmid, expressed, purified, and finally biotinylated for SPR analysis. Nearly 200 drug fragments were screened in-vitro for affinity to the AVI-FERM protein using SPR. Results: Nearly 200 drug fragments were screened for binding resulting in 23 molecules with visibly slow dissociation curves. Eight of those molecules meet the second “hit” criteria of RUmax greater than 8 RU. Conclusions: The protein construct prepared was successful in its first fragment screening experiment and will prove to be useful in further experimenting the hit fragments and also in further experiments on FERM drug discovery. | en_US |
